چکیده

Radioactively labeled thymidine (TdR) has been used extensively to measure bacterial production in aquatic environments, but critical assumptions of the TdR technique often have gone untested. In this study of a coastal marine sediment, the metabolic fate of methyl [3H]TdR and methyl [14C]TdR was at variance with the assumptions necessary for determining bacterial production. Only 2% of incorporated radioactivity was recovered in the DNA fraction of TCA-insoluble material following time-course incubations of l-300 min. At least the methyl group of TdR was extensively catabolized, as shown by copious production of 14C0,. The temporal patterns of 3H : 14C ratios in macromolecuAcknowledgments We thank P. LaRock). lar fractions indicated that products of catabolism were recycled into the DNA fraction. The accuracy of the TdR technique depends in large part on the degree to which such catabolism occurs. Estimation of bacterial production in aquatic environments is of interest to microbial ecologists, organic geochemists, limnologists, and oceanographers, among others. There has been a recent proliferation of such estimates, accompanied by controversy regarding the technique of choice (e.g. Hagstriim et al. 1979; Karl 1986; Moriarty 1986; Fuhrman et al. 1986; Karl and Winn 1986). Many investigators use radiolabeled precursors of nucleic acids and, for example, incorporation of radioactivity into deoxyribonucleic acid (DNA) as a measure of bacterial growth. Undoubtedly the most popular precursor has been the nucleoside thymidine (thymidine deoxyribose, hereafter TdR); methyl [3H]TdR is the most frequently used form. The expectation has been

Bacterial production in marine sediments: will cell-specific measures agree with whole-system metabolism?

Estimates for bacterial production in nearshore western Atlantic Ocean sediments were made using the frequency of dividing cell and thymidine uptake methods. The frequency of dividing cell (FDC) method resulted in estimates in the range of 5 to 50 g C m-' d-'. The thymidine uptake (THY) method resulted in estimates of 1 to .8 g C mP2 d-'. Metabolic measurements based on sediment oxygen uptake s...

Isolation of L-Glutaminase Producing Marine Actinomycetes

L Glutaminase, a therapeutic enzyme obtained from marine Actinomycetes gains importance because of their adaptability to varied environmental factors. An experimental study was carried out to isolate the L glutaminase producing actinomycetes from the marine sediment of Thoothukudi. Marine sediment sample was collected from six different locations of the Thoothukudi coastal ecosystem, enriched w...

Calibration method of numerical models in coastal sediment studies of Kuhmobarak area

Abstract Application of numerical models is useful in coastal sediment studies; however, it is essential to calibrate the models in an appropriate method. Empirical equations, historical satellite imagery and other coastal data and evidence are proposed for the models calibration in the vicinity of coastal indicators around the site of projects. In this study, coastal sediment process of K...

A study on the composition of size-fractionated suspended particulate matter in shallow coastal waters of Chabahar Bay

Abstract In the present study, sampling and analysis of size-fractionated suspended particulate matter (SPM) in coastal waters of Chabahar Bay have been done for the first time. Sampling has been conducted on December 25 of 2018 from 5 stations in different locations of the Bay in order to evaluate the effects of natural and human activities on SPM mass and composition. With an overall average...

Production of planktonic bacteria in Lake Michigan’

Bacterial production rates were estimated for the surface waters of a station 100-m deep in southeastern Lake Michigan during 1984. Production was calculated from incorporation of C3Hmethyllthymidine and from empirical conversion factors determined from dilution experiments performed throughout the study. The conversion factors (with typical C.V. ~40%) varied between 4.7 and 18.3 x lo9 cells pr...
